Solid state screenings
Theses studies allow to gather knowledge on the nature of the solid states of a give compound, either intermediate or Active Pharmaceutical Ingredient (API), and its related properties and thus select the right solid phases.
Reliable solid state studies are an essential prerequisite for robust crystallization process and pharmaceutical development, and the elaboration of IP, patents and regulatory files.
Holodiag specializes in solid state screenings, which are specific investigations on the nature of a compound’s solid states. We perform numerous experiments (1000+) on small-scale samples of a few mg, controlled by X-Ray Powder Diffraction (XRPD) to highlight new solid states. We carry out specific screenings for polymorphs/hydrates/solvates, salts, co-crystals and amorphous solid dispersions.
Once a solid state is highlighted, our scientists proceed to the investigation of physico-chemical properties.
Holodiag offers solid state screenings and studies of polymorphs. We help pharmaceutical customers control the polymorphism of their Active Pharmaceutical Ingredient (API) and study various crystallization conditions for the identification of new phases.
Polymorphism is the ability for a molecule to crystallize under several solid forms. Crystallization screening of polymorphic forms or solvates gives the opportunity to identify a maximum number of solid phases. The control of polymorphism thus avoids late stage development problems du to the appearance of new phases. This also secures customer’s Intellectual Property (IP).
Salt screening is a step that cannot be overlooked. Salt screenings imply salt formation with counter-ions and crystallization in solvents. Salts enhance the solubility of APIs, can make up for amorphous solid obtention and broaden the portfolio of solid phases, and thus secure customer’s intellectual property.
Co-crystals screening should be considered when access to new physico-chemical characteristics is required and polymorphism and salt-formation have already been studied.
We have the expertise to choose the right co-crystallizer according to specific interactions that are related to the structure of the molecule (H-bonds, pi-pi interactions, Van der Waals, etc.).
Amorphous solid dispersions
Holodiag conducts specific tests to gain access to amorphous Active Pharmaceutical Ingredients (APIs) within a matrix. This enables access to enhanced properties such as dissolution rate and solubility, and thus increased bioavailability.
